El Populo lies in the upper Florida Keys near Islandia, in the patch reef and shallow reef habitat typical of the Biscayne National Park area on the Atlantic side of Key Largo. Sites in this stretch of the Keys are generally accessed by boat out of nearby marinas in Key Largo or Homestead.
